i'm building a paver patio, but there is a very thick wisteria root (about 3inches thick) smack on the edge of where i want to finish the paver patio. Will my digging the dirt around it (the root) kill it? If i place paver base material around it, will it kill the plant, or on it directly in order to finish my patio do i run the risk that the plant will push through the paver and ruin the patio?

You do run the risk of the roots moving the pavers after they've been laid - a large plant like that near the edge of a paved area means there's a strong likelihood of roots, at some point, causing the paving to shift, lift and tip. Usual procedure where the roots are present of a large plant or tree is to use different surfacing in that area, such as gravel, pebbles, etc.
